Microbial Natural Products

Prof Dr Rolf Müller

Natural products of microbial origin continue to be a promising source for the development of novel pharmaceuticals. Our department focuses on the discovery, as well as the biotechnological engineering and production of myxobacterial natural products with antimicrobial properties.

Our research and approach

Myxobacteria are capable of producing a plenitude of natural products with high structural and functional diversity. Accordingly, we find myxobacterial substances acting against all kinds of organisms, ranging from bacteria and fungi to mammalian cells or even viruses. We apply activity- and structure-guided approaches to identify bioactive compounds, but also use modern genome mining tools to shed light on the hidden biosynthetic potential of underexplored myxobacteria. Following their identification, we structurally characterize novel compounds and solve the molecular mechanisms underlying their biosynthesis and mode of action. This provides vital information for the biotechnological and chemical improvement of these substances. Our goal is to generate and develop hit and lead compounds, which are eventually transferred to the clinic. We have successfully established an in-house strain collection containing more than 10.000 unique myxobacterial strains, isolated from places all over the world. This collection is continuously expanded and represents the basis of our pursuit for exciting novel chemistry.

Isolation of New Myxobacteria for Anti-infective Research

Inspired by the diversity of uncultured myxobacteria found by metagenomics, our isolation strategy focuses on bringing these microorganisms to cultivations in the laboratory. The search for these bacteria is boundless to types of samples, environments, and geographical locations. However, we are especially interested in under-explored environments (e.g. oceans) and as well as those habitats with high microbial diversity and interactions to compete for nutrients and territoriality (e.g. forest soils). Myxobacteria are an outstanding source of bioactive compounds. Our previous studies have shown that phylogenetically distant myxobacterial taxa are especially likely to produce novel compounds. Thus, our isolation is aimed towards the “uncultured” taxa, which harbour a high potential for the discovery of novel anti-infectives. Culture conditions in the laboratory are adjusted to the sample to support the growth of contained myxobacteria and allow for the production of secondary metabolites. Successfully isolated strains are subjected to intense physiological characterization, growth optimization, and fermentation to screen these strains for bioactive compounds. We also perform genome sequencing to correlate the compounds with the respective genes and unveil further metabolites hidden in the genome. In the last two years, we also started exploring different regions of Saarland for novel myxobacteria. The local public is invited to assist HIPS scientists with collecting soil samples via the citizen science project “Sample’ das Saarland”
