Drug Delivery Across Biological Barriers

Prof Dr Claus-Michael Lehr

The research focus of Prof. Lehr’s group is on new technologies for the safe and efficient delivery of anti-infectives across biological barriers, such as the lungs, the intestinal mucosa and the skin, but also mucus, biofilms and the bacterial cell envelope.

Our research and approach

Advances in molecular biotechnology and medicinal chemistry are continuously generating new drug candidates. However, developing these molecules into successful therapeutics also requires optimizing their biopharmaceutical properties, in particular the ability to cross biological barriers, as represented by tissues, cells and cellular membranes. As further optimizing the active compounds is often not possible, new technologies are needed to ensure safe and effective delivery to the site of action, ideally by non-invasive (“needle-free”) administration. In this context we are working both on innovative drug carriers as well as on predictive in vitro models with the aim to accelerate preclinical development and to facilitate clinical translation.

LUNG-ON-A-CHIP: Bioengineered microfluidic in vitro test system to study alveolar transport and clearance

The aim of the project focuses on the design and integration of an innovative bioengineered microfluidic system that mimics the characteristic physiological microenvironment as well as the unique cellular composition of the lung. By reconstituting the human alveolar epithelial barrier in vitro, our organ-on-chip systems serve as an integrative platform for in vitro analysis of both, transport across the air-blood-barrier into the blood stream as well as uptake and subsequent clearance by alveolar macrophages. Such model is expected to have significant impact for predictive preclinical characterisation of advanced aerosol medicines, in healthy as well as in diseased state.

ANTI-TB: Antibiotika-beladene Nanocarrier zur Therapeutischen Inhalation gegen Tuberkulose

The ANTI-TB project aims to develop a novel therapy for tuberculosis. Tuberculosis is the world's most serious bacterial infection in humans. Their control is hampered by the rising occurrence of therapy-resistant M tuberculosis strains. The project uses a nanomedical approach to improve the unsatisfying therapy situation, in particular for infections with therapy-resistant pathogens, and to reduce further resistance developments. Nanocarriers will be combined with new anti-tuberculosis drugs and optimized with specific surface modifications that direct them to infection sites, infected cells, and pathogens. For ease of use, the most effective antibiotic nanocarriers will ultimately be converted into an aerosol or powder inhalation therapy. The strategy aims to increase the local drug concentrations in the lung and shorten the therapy duration. Such an approach should minimize the onset of drug-resistant pathogens, reduce systemic side effects, and improve patients' preparedness for therapy. The HIPS investigates the effects and interactions of the nanoparticles with the lungs' physiological fluids, mucus and lung surfactant, as well as macrophages and epithelial cells. The aerosol powder generation is also our task.

NEW DEAL: New nanotherapy research for inflammatory bowel diseases

The NEW DEAL project aims to develop a highly selective and local therapy with an improved efficacy and safety profile for IBD patients. Specific siRNAs targeting Jak-1 or Jak-3, nanoparticle carriers, and formulations for oral application are developed. This treatment might be an alternative to the existing therapies. In this project, the HIPS is contributing with its expertise on advanced coculture models of the inflamed intestinal barrier and the mucus. We investigate the intestinal barrier crossing of the nanocarriers.
